What Allogene Therapeutics, Inc. does

In the company’s own words

Allogene Therapeutics, Inc. (NASDAQ: ALLO) is a clinical-stage immuno-oncology company pioneering the development of genetically engineered allogeneic T cell product candidates for treating cancer and autoimmune diseases. The company's website is www.allogene.com.

The company develops off-the-shelf T cell therapies derived from healthy donors for use in any eligible patient, rather than autologous approaches requiring individual patient cell collection and manufacturing. This allogeneic strategy aims to deliver treatments faster, more reliably, and at greater scale than traditional autologous therapies. Allogene's approach involves engineering T cells to minimize graft-versus-host disease risk through genetic modifications that eliminate functional T cell receptors, while incorporating chimeric antigen receptors designed to target specific cancer cells or pathogenic immune cells.

Allogene's pipeline focuses on three core programs. Cemacabtagene ansegedleucel, or cema-cel, targets CD19 for large B-cell lymphoma treatment and is being evaluated in the pivotal Phase 2 ALPHA3 trial as a first-line consolidation therapy for newly diagnosed patients with minimal residual disease. ALLO-316 targets CD70 for renal cell carcinoma and received Regenerative Medicine Advanced Therapy designation in 2024. ALLO-329, targeting both CD19 and CD70, is in development for autoimmune diseases including systemic lupus erythematosus, idiopathic inflammatory myopathies, and systemic sclerosis.

The company operates Cell Forge 1, a current good manufacturing practices facility in Newark, California, for manufacturing product candidates used in clinical studies. Allogene has established strategic partnerships including an exclusive worldwide oncology license from Cellectis for TALEN gene-editing technology, a non-exclusive license from Arbor Biotechnologies for CRISPR-based gene-editing technology, and an exclusive license and collaboration agreement with Servier for developing and commercializing cema-cel in the United States, European Union, and United Kingdom.

Founded with leadership from executives previously instrumental in developing Yescarta at Kite Pharma, Allogene has treated more than 200 patients across six clinical studies. The company anticipates multiple clinical readouts in the second quarter of 2026 that could validate key scientific assumptions underlying off-the-shelf CAR T therapy.
