Cogent Biosciences, Inc. (NASDAQ: COGT) is a clinical-stage biotechnology company focused on developing precision therapies for genetically defined diseases. The company's approach centers on designing rational precision medicines that target the underlying genetic causes of disease to improve patient outcomes.
The company's most advanced program is bezuclastinib, also known as CGT9486, a highly selective tyrosine kinase inhibitor designed to potently inhibit the KIT D816V mutation and other mutations in KIT exon 17. Bezuclastinib is being developed to treat patients with Systemic Mastocytosis, including both Non-Advanced Systemic Mastocytosis and Advanced Systemic Mastocytosis, as well as gastrointestinal stromal tumors. In 2025, Cogent reported positive top-line results from registrational trials in each of these indications and submitted its first New Drug Application for bezuclastinib in patients with Non-Advanced Systemic Mastocytosis in December 2025. The company expects to launch bezuclastinib commercially in the United States in the second half of 2026, pending regulatory approval.
Beyond bezuclastinib, Cogent is building a pipeline of additional precision therapies. The company has an ongoing Phase 1 study of CGT4859, a novel FGFR2/3 inhibitor, and initiated a Phase 1 study of CGT4255, a CNS-penetrant, selective mutant ErbB2 inhibitor. The company is also developing CGT6297, a PI3Kα inhibitor, with Phase 1 initiation expected in the first quarter of 2026. The Cogent Research Team, based in Boulder, Colorado, is advancing preclinical programs targeting KRAS and JAK2 mutations, with plans to submit investigational new drug applications for these programs in 2026.
Cogent has assembled a management team with extensive experience in research, development, manufacturing, and commercialization of pharmaceutical products, particularly precision medicines for genetically defined diseases. The company is building an internal commercial organization to support the anticipated launch of bezuclastinib. More information about the company is available at www.cogentbio.com.
