What MADRIGAL PHARMACEUTICALS, INC. does
In the company’s own words
Madrigal Pharmaceuticals, Inc. (MDGL: Nasdaq) is a biopharmaceutical company focused on developing and commercializing novel therapeutics for metabolic dysfunction-associated steatohepatitis, a serious liver disease characterized by inflammation, cellular damage, and fibrosis that can progress to cirrhosis, liver failure, and liver cancer. The company's website is www.madrigalpharma.com.
Madrigal's primary product is Rezdiffra (resmetirom), a once-daily oral liver-directed thyroid hormone receptor beta agonist approved by the FDA in March 2024 for adults with noncirrhotic MASH with moderate to advanced liver fibrosis. The European Commission granted conditional marketing authorization in August 2025, making Rezdiffra the first medication approved by both regulatory bodies for this indication. The company launched Rezdiffra in the United States in April 2024 and in Germany in September 2025, generating $958.4 million in product revenue during 2025.
Beyond its approved therapy, Madrigal is building a focused pipeline to establish industry leadership in MASH treatment. The company has acquired rights to MGL-2086, an oral glucagon-like peptide-1 receptor agonist; ervogastat, an oral DGAT2 inhibitor; and six small interfering RNA programs designed to target genes implicated in MASH disease progression. These candidates are being evaluated for potential combination therapy with Rezdiffra to enhance efficacy across the MASH spectrum.
The company is advancing two significant clinical trials to expand Rezdiffra's market opportunity. The MAESTRO-NASH OUTCOMES trial, which completed enrollment of 845 patients, is evaluating Rezdiffra in patients with compensated MASH cirrhosis, with results expected in 2027. Positive outcomes could potentially double Rezdiffra's commercial opportunity by supporting approval in this patient population with no currently available treatments. Additionally, the company expects outcomes data from its MAESTRO-NASH trial in 2028 to support full FDA approval.
Madrigal operates under an exclusive worldwide license agreement with Hoffmann-La Roche for resmetirom development and commercialization rights. The company secured $350 million in senior secured financing in July 2025 with access to additional delayed draw and incremental facilities totaling up to $400 million.
