What Septerna, Inc. does
In the company’s own words
Septerna, Inc. (Nasdaq: SEPT) is a clinical-stage biotechnology company pioneering G protein-coupled receptor (GPCR) oral small molecule drug discovery through its proprietary Native Complex Platform. The company's website is www.Septerna.com.
The company focuses on developing a deep pipeline of oral small molecule GPCR-targeted programs across three initial therapeutic areas: endocrinology, immunology and inflammation, and metabolic diseases, with plans to expand into neurology, women's health, cardiovascular, and respiratory disease. Septerna's Native Complex Platform represents a foundational innovation that enables the isolation, purification, and reconstitution of full-length GPCR proteins within ternary complexes with ligands and transducer proteins in a lipid bilayer environment that mimics the cell membrane. This approach allows the company to apply state-of-the-art discovery tools including structure-based drug design, computational docking, and DNA-encoded libraries to identify and optimize novel small molecule candidates.
The company's lead programs include SEP-479, an oral small molecule PTH1R agonist for hypoparathyroidism, which is planned to enter Phase 1 clinical trials in the first half of 2026, and SEP-631, a selective oral small molecule MRGPRX2 negative allosteric modulator for chronic spontaneous urticaria, which completed Phase 1 trials with positive results announced in March 2026. Septerna has also established a strategic partnership with Novo Nordisk A/S focused on developing oral small molecule therapies for metabolic-related diseases targeting GLP-1, GIP, and glucagon receptors.
GPCRs represent the largest and most diverse family of cell membrane receptors and account for approximately one-third of all FDA-approved drugs, representing approximately 500 products with combined global revenue of approximately $125 billion in 2023. Despite this success, approximately 75 percent of potential GPCR therapeutic targets remain undrugged, representing significant opportunity for novel therapeutic development. Septerna's platform is designed to address this gap by enabling the discovery of oral small molecules targeting previously intractable GPCR targets and novel binding pockets on validated receptors.
