Spyre Therapeutics, Inc. The company's pipeline includes extended half-life antibodies targeting α4β7, TL1A, and IL-23 in development as monotherapies and pair-wise combinations designed to enable quarterly or twice-annual subcutaneous maintenance dosing.
The company's lead program, SPY001, is a highly potent and selective anti-α4β7 monoclonal antibody in Phase 2 development for ulcerative colitis and Crohn's disease. SPY002 and SPY072 are anti-TL1A antibodies being developed for inflammatory bowel disease and rheumatic diseases respectively, while SPY003 is an anti-IL-23 antibody for IBD. Spyre is also advancing four combination programs: SPY120 combining anti-α4β7 and anti-TL1A mechanisms, SPY130 combining anti-α4β7 and anti-IL-23, and SPY230 combining anti-TL1A and anti-IL-23.

Spyre's intellectual property portfolio includes multiple patent families covering compositions of matter, methods of use, and pharmaceutical formulations across its programs, with expected patent expirations ranging from 2044 to 2046. The company licenses its core technology from Paragon Therapeutics pursuant to exclusive worldwide license agreements and outsources all manufacturing and supply activities to third-party manufacturers including WuXi Biologics. As of December 31, 2025, Spyre employed 102 full-time employees. The company was formed through the acquisition of Spyre Therapeutics' assets by Aeglea BioTherapeutics, which subsequently changed its name to Spyre Therapeutics in November 2023.
