Aligos Therapeutics Inc. (Nasdaq Capital Market: ALGS) is a clinical-stage biotechnology company focused on developing novel therapeutics to address unmet medical needs in liver diseases and viral infections. The company's pipeline includes three potential best-in-class drug candidates targeting chronic hepatitis B virus infection, metabolic dysfunction-associated steatohepatitis and obesity, and coronavirus infections.
The company's lead program, pevifoscorvir sodium, is a capsid assembly modulator designed for chronic HBV infection. In Phase 1 clinical trials spanning up to 96 weeks, pevifoscorvir sodium demonstrated multi-log10 reductions in viral antigens and substantial HBV DNA suppression compared to standard nucleos(t)ide analog treatments. The company is currently advancing this candidate in the Phase 2 B-Supreme study, a randomized, double-blind, active-controlled trial evaluating safety and efficacy in approximately 200 untreated subjects with chronic HBV infection, with topline data expected in 2027.
ALG-055009, a thyroid hormone receptor beta agonist, is being developed for MASH and obesity. Phase 2a data demonstrated statistically significant reductions in liver fat content at 12 weeks in subjects with presumed MASH. Preclinical data suggests synergistic fat mass loss when combined with incretin receptor agonists. The company is evaluating various funding options for continued development, including potential out-licensing.
ALG-097558 is a ritonavir-free pan-coronavirus protease inhibitor showing threefold greater potency than approved coronavirus protease inhibitors in cell-based assays. Phase 1 clinical studies demonstrated favorable tolerability and pharmacokinetics without requiring ritonavir boosting. The candidate is currently in Phase 2 clinical studies sponsored by AGILE University of Liverpool in standard and high-risk COVID-19 patients.
The company's management team brings decades of combined experience in drug discovery and development for viral infections and liver diseases, with involvement in the discovery and development of multiple commercially successful drugs. Aligos maintains internal capabilities in small molecule and oligonucleotide chemistry, with dedicated chemistry, manufacturing, and control teams supporting contract development and manufacturing organizations.
