The company's lead product candidate, buntanetap, is an orally administered, brain-penetrant small molecule designed to inhibit the synthesis of neurotoxic proteins including APP/Aβ, tau/phospho-tau, α-Synuclein, and TDP43. These proteins are believed to be primary drivers of neurodegeneration. Buntanetap operates through a novel mechanism of action as a translational inhibitor, targeting an atypical stem loop region in the messenger RNA of neurotoxic aggregating proteins. By preventing overexpression of these proteins, the drug is designed to reverse the downstream toxic cascade that leads to neurodegeneration and restore nerve cell function.
Annovis has completed twelve clinical studies to date, including Phase 1/2 trials in early Alzheimer's and Parkinson's patients and a Phase 2/3 trial in mild to moderate Alzheimer's patients. The company initiated a pivotal Phase 3 trial in early Alzheimer's disease in February 2025, enrolling 760 patients across 83 U.S. sites. The trial incorporates a six-month symptomatic efficacy component followed by a twelve-month extension to evaluate disease-modifying potential. Additionally, the company began an open-label extension study in Parkinson's disease patients in January 2026, designed to enroll 500 patients for long-term safety and efficacy evaluation.
