Aura's lead candidate, belzupacap sarotalocan (bel-sar), represents a novel class of drugs called virus-like drug conjugates (VDCs) that employ a dual mechanism of action. The therapeutic approach combines acute tumor cell killing through light-activated cytotoxic payload delivery with activation of secondary immune-mediated responses that generate long-term antitumor immunity. Bel-sar consists of modified human papillomavirus-derived capsid proteins conjugated to light-activated molecules, enabling precise tumor cell elimination with minimal damage to surrounding healthy tissue.

The company is advancing bel-sar through late-stage clinical development across multiple oncology indications. In ocular oncology, bel-sar is in a global Phase 3 trial for early choroidal melanoma, the most common primary malignant intraocular tumor affecting approximately 11,000 patients annually in the United States and Europe. The company has also received Orphan Drug Designation and Fast Track Designation from the FDA for this indication. Additionally, bel-sar is in Phase 2 development for metastases to the choroid and Phase 1 proof-of-concept testing for cancers of the ocular surface.

In urologic oncology, Aura is developing bel-sar for non-muscle-invasive bladder cancer (NMIBC), which represents a significant unmet medical need affecting approximately 80,000 patients annually in the United States.
