Since 2012, Corcept has marketed Korlym (mifepristone) 300 mg tablets in the United States for treating patients with hypercortisolism, also known as Cushing's syndrome. In June 2024, the company introduced an authorized generic version of Korlym for the same indication. Korlym works by competing with cortisol's binding to the glucocorticoid receptor, thereby modulating the effects of abnormal cortisol levels without compromising cortisol's healthy functions. However, mifepristone also binds to the progesterone receptor, causing adverse effects including endometrial thickening and vaginal bleeding in a significant portion of patients.
The company has discovered more than 1,000 proprietary selective cortisol modulators that bind to the glucocorticoid receptor without affinity for the progesterone receptor, eliminating pregnancy termination and related adverse effects. Lead compounds include relacorilant, nenocorilant, miricorilant, and dazucorilant, which are being evaluated in clinical trials for hypercortisolism, various solid tumors including ovarian, endometrial, cervical, pancreatic and prostate cancers, amyotrophic lateral sclerosis, and metabolic dysfunction-associated steatohepatitis.
Corcept submitted a New Drug Application for relacorilant to treat endogenous hypercortisolism in December 2024, though the FDA issued a Complete Response Letter in December 2025 requesting additional efficacy evidence. The company has also submitted an NDA for relacorilant combined with nab-paclitaxel for platinum-resistant ovarian cancer, with a PDUFA date of July 11, 2026. Clinical trial results have demonstrated clinically meaningful improvements in blood pressure, glucose control, weight, and body composition across multiple indications.
The company relies on contract manufacturers for production and holds significant quantities of the active pharmaceutical ingredient. Research and development expenses totaled $254.9 million in 2025, representing 36 percent of total operating expenses. Corcept's intellectual property portfolio includes U.S. and foreign patents covering its selective cortisol modulators with expiration dates ranging from 2028 to 2042.
