The company conducts its primary banking operations through Busey Bank, an Illinois state-chartered bank founded in 1868, which provides comprehensive financial services to consumers and businesses across ten states. Busey Bank maintains 79 banking centers strategically located throughout Illinois, Missouri, Texas, Colorado, Florida, Kansas, Oklahoma, Arizona, Indiana, and New Mexico, with the largest concentration of 50 locations in Illinois.

The company's core business segments include traditional banking services, wealth management, and payment technology solutions. Busey Bank offers commercial and industrial lending, commercial real estate financing, real estate construction loans, agricultural loans, residential mortgage products, consumer loans, and deposit services. The wealth management division provides asset management, investment advisory, trust and estate services, financial planning, and farm management to individuals, businesses, and foundations, with $15.66 billion in assets under care as of December 31, 2025. FirsTech, a wholly owned subsidiary, delivers technology-enabled payment processing solutions including mobile bill payment, electronic payment routing, lockbox processing, and merchant services across multiple industries.

Busey operates through five geographic regions—East, Midwest, Central, Texas, and West—and maintains industry verticals including Life Equity Lending, Sponsor Finance, Energy Lending, and SBA Lending. The company employs 1,914 full-time equivalents across its footprint. As of June 30, 2025, Busey Bank ranked fourth in total deposits among 349 financial institutions headquartered in Illinois and maintains top-ten market share rankings in multiple counties across its operating regions.

The company's growth strategy has been supported by strategic acquisitions, including the March 2025 acquisition of CrossFirst Bankshares and the April 2024 acquisition of Merchants and Manufacturers Bank Corporation.
