What Korro Bio, Inc. does

In the company’s own words

Korro Bio, Inc. (NASDAQ: KRRO) is a biopharmaceutical company focused on discovering, developing, and commercializing genetic medicines based on RNA editing technology. The company's website is www.korrobio.com.

Korro Bio was formed through a November 2023 business combination between Frequency Therapeutics, Inc. and privately held Korro Bio Ops, Inc. The company operates a proprietary platform called OPERA (Oligonucleotide Promoted Editing of RNA) that harnesses the body's natural RNA editing process to make precise, transient changes to RNA nucleosides, specifically converting adenosine to inosine edits. This approach differs from DNA editing by offering improved specificity, reduced off-target effects, and the ability to re-dose patients, while leveraging established oligonucleotide manufacturing and delivery technologies.

The company's pipeline includes four wholly-owned programs and up to two programs under a collaboration with Novo Nordisk that entered a 12-month pause in November 2025. The lead program, KRRO-121, targets hyperammonemia in patients with urea cycle disorders and hepatic encephalopathy through enhanced ammonia clearance via glutamine synthetase stabilization. The company anticipates a regulatory filing for KRRO-121 in the second half of 2026. Additional programs include a next-generation GalNAc-conjugated therapy for alpha-1 antitrypsin deficiency with development candidate nomination expected in the second quarter of 2026, a longevity and liver health program targeting AMPKγ1 activation, and an ALS program targeting TDP-43 modification.

Korro Bio's OPERA platform integrates expertise in ADAR biology, oligonucleotide chemistry, machine learning optimization, and established delivery modalities including GalNAc-conjugated delivery for subcutaneous administration. The company leverages precedented regulatory pathways for oligonucleotide medicines and can design and synthesize approximately 1,200 oligonucleotides monthly with up to 6,000 assay data points per target. All programs remain in research or preclinical development stages. The company has cash runway extending into the second half of 2028 to support advancement of its pipeline and achievement of anticipated milestones.
