In the company’s own words
LeonaBio, Inc. (Nasdaq Capital Market: LEON) is a clinical-stage biopharmaceutical company dedicated to developing novel therapeutics for high unmet medical needs, particularly in oncology and neurology. The company's website is www.leonabio.com.
LeonaBio operates a pipeline of drug candidates focused on two primary therapeutic areas. The company's lead oncology asset is lasofoxifene, a selective estrogen receptor modulator currently in Phase 3 clinical development for the treatment of estrogen receptor-positive, HER2-negative metastatic breast cancer with ESR1 mutations. Lasofoxifene was in-licensed globally in December 2025, excluding Asia and certain Middle Eastern countries. The company's pivotal ELAINE-3 trial aims to demonstrate the superiority of lasofoxifene combined with abemaciclib compared to fulvestrant with abemaciclib for patients with ESR1-driven metastatic breast cancer following CDK4/6 inhibitor progression. As of December 2025, the trial had completed over fifty percent recruitment, with topline data anticipated in the second half of 2027.
In neurology, LeonaBio's lead candidate is ATH-1105, a small-molecule positive modulator of the neurotrophic HGF system designed to treat amyotrophic lateral sclerosis. The company completed a first-in-human Phase 1 trial in November 2024, which demonstrated favorable safety and tolerability in eighty healthy volunteers. A Phase 2 proof-of-concept trial in ALS patients is anticipated to commence in 2026, with primary focus on plasma neurofilament light chain levels as a biomarker of neurodegeneration.
The company also maintains early-stage development programs including ATH-1020, another HGF system modulator being explored for neuropathic pain and other neurological indications, along with novel compounds in discovery targeting various molecular pathways relevant to oncology and neurology applications.
LeonaBio has incurred significant losses since inception and anticipates continued losses through the foreseeable future as it advances clinical development programs. The company is classified as a smaller reporting company and is not a well-known seasoned issuer. As of March 27, 2026, the company had 9,393,514 shares of common stock outstanding.
