MacroGenics, Inc. (NASDAQ: MGRC) is a clinical-stage biopharmaceutical company focused on developing innovative antibody-based therapeutics for cancer treatment. The company generates its pipeline of product candidates from proprietary antibody technology platforms, including antibody-drug conjugates (ADCs) and multi-specific antibodies utilizing its DART and TRIDENT platforms. MacroGenics maintains a commercial-scale cGMP antibody manufacturing facility at its Maryland headquarters, which supports both internal clinical programs and provides outsourced contract development and manufacturing services to collaborators and third parties.

The company's wholly-owned clinical pipeline includes three primary product candidates: lorigerlimab, a bispecific DART molecule targeting PD-1 and CTLA-4 checkpoint inhibitors; MGC026, an ADC targeting B7-H3; and MGC028, a first-in-class ADC targeting ADAM9. MacroGenics also maintains economic rights to three FDA-approved products: TZIELD (teplizumab-mzwv) for type 1 diabetes, ZYNYZ (retifanlimab-dlwr) for Merkel cell carcinoma and squamous cell carcinoma of the anal canal, and MARGENZA (margetuximab-cmkb) for HER2-positive breast cancer, which was sold to TerSera Therapeutics in November 2024.

Strategic collaborations represent a significant component of MacroGenics' business model. The company has generated over $1.6 billion in non-dilutive funding since inception through partnerships with global biopharmaceutical companies. A notable collaboration with Gilead Sciences involves MGD024, a next-generation CD123 × CD3 DART molecule for hematologic malignancies, with potential milestone payments reaching approximately $1.6 billion across three programs.
