The company's lead asset is ateganosine, an investigational dual mechanism of action drug candidate that incorporates telomere targeting and immunogenicity. Ateganosine works by targeting telomeres, the repetitive DNA sequences at chromosome ends that play a fundamental role in cancer cell survival and resistance to current therapies. The drug's mechanism involves telomerase recognition of ateganosine's metabolite, which becomes incorporated into cancer cell telomeres, creating faulty structures that break apart and trigger cancer cell death. The company believes ateganosine transforms immunologically "cold" tumors into "hot" tumors responsive to immunotherapy, a process occurring within 24 to 72 hours.

MAIA is advancing ateganosine through multiple clinical programs. The Phase 2 THIO-101 trial evaluates ateganosine in sequential combination with checkpoint inhibitors for non-small cell lung cancer patients. In 2025, the company initiated the Phase 3 THIO-104 pivotal trial comparing ateganosine sequenced with checkpoint inhibitors to chemotherapy in third-line NSCLC patients. Additional Phase 2 trials are planned for hepatocellular carcinoma, colorectal cancer, and small cell lung cancer in combination with checkpoint inhibitors.

The company has established strategic collaborations with major pharmaceutical partners including Regeneron for cemiplimab supply, BeOne Medicines for tislelizumab, and Roche for atezolizumab. MAIA has received multiple regulatory designations including orphan drug designations for hepatocellular carcinoma, small cell lung cancer, and malignant gliomas, as well as rare pediatric disease designation and fast track designation for NSCLC treatment.

MAIA maintains operations in Chicago, Illinois, with a wholly-owned Australian subsidiary established in 2021 and a Romanian subsidiary established in 2022. The company is developing a pipeline of second-generation telomere-targeting compounds, with seven molecules selected for further evaluation in preclinical and in vivo models.
