What NextCure, Inc. does
In the company’s own words
NextCure, Inc. (Nasdaq: NXTC) is a clinical-stage biopharmaceutical company focused on developing innovative medicines for cancer patients who do not respond to or have disease progression on current therapies. The company specializes in antibody-drug conjugates, or ADCs, which consist of monoclonal antibodies conjugated to cytotoxic drugs via chemical linkers. NextCure's core competency lies in understanding biological pathways, biomarkers, and cellular interactions within the tumor microenvironment to design targeted cancer therapies.
The company's lead product candidate is SIM0505, a novel ADC directed to cadherin-6, or CDH6, featuring a proprietary topoisomerase 1 inhibitor payload. Developed by partner Simcere Zaiming Pharmaceutical Co., Ltd., SIM0505 targets multiple solid tumors including ovarian, endometrial, non-small cell lung, and renal cancers. NextCure obtained exclusive worldwide rights to develop, manufacture, and commercialize SIM0505 outside of China, Hong Kong, Macau, and Taiwan. The company dosed its first patient in a Phase 1 clinical trial in October 2025 and expects to provide dose escalation data in the second quarter of 2026.
The company's second major program is LNCB74, a B7-H4 targeted ADC being developed under a cost-sharing collaboration with LigaChem Biosciences, Inc. LNCB74 combines a NextCure-generated monoclonal antibody with LigaChem's proprietary linker technology and targets breast, ovarian, and endometrial cancers. The first patient was dosed in January 2025, with a trial update expected in the second half of 2026.
NextCure maintains a dedicated, state-of-the-art cGMP manufacturing facility with 2,000-liter production capacity, though internal manufacturing operations were paused in March 2024 as part of a strategic restructuring. The company also pursues partnerships for clinical programs NC410 and NC525, as well as preclinical non-oncology programs including NC181 for Alzheimer's disease and NC605 for osteogenesis imperfecta.
