What PMV Pharmaceuticals, Inc. does

In the company’s own words

PMV Pharma, Inc. (NASDAQ: PMVP) is a precision oncology company pioneering the discovery and development of small molecule therapies targeting p53 mutations and other p53-related cancers. The company's website is www.pmvpharma.com.

PMV Pharma leverages more than four decades of p53 research experience, building upon foundational work by co-founder Dr. Arnold Levine who discovered the p53 protein in 1979. The company has developed a precision oncology platform designed to generate selective, small molecule, tumor-agnostic therapies that structurally correct specific mutant p53 proteins to restore their wild-type function. P53 mutations are found in approximately half of all human cancers, representing a substantial opportunity for targeted treatment approaches.

The company's lead product candidate, rezatapopt, is an orally available small molecule designed to potently and selectively correct p53 misfolding caused by the Y220C mutation while sparing wild-type p53. The p53 Y220C mutation is associated with approximately one percent of all cancers, including endometrial, breast, non-small cell lung cancer, colorectal, pancreatic, and notably approximately three percent of ovarian cancers. Rezatapopt received FDA Fast Track designation in October 2020 and orphan drug designation in March 2026 for treatment of TP53 Y220C positive ovarian cancer, fallopian tube cancer, and primary peritoneal cancer.

PMV Pharma initiated the Phase 1/2 PYNNACLE clinical trial for rezatapopt in October 2020, with the first patient dosed in the pivotal Phase 2 monotherapy portion in the first quarter of 2024. The company announced interim Phase 2 data in September 2025, updated in October 2025, demonstrating an overall response rate of thirty-four percent across eight tumor types and forty-six percent in the ovarian cancer cohort. The company plans to submit a New Drug Application to the FDA in the first quarter of 2027 for treatment of patients with platinum-resistant or refractory ovarian cancer harboring the TP53 Y220C mutation.

Beyond rezatapopt, PMV Pharma is developing a pipeline of additional product candidates targeting other p53 mutations using its precision oncology platform. The company does not operate manufacturing facilities and relies on third-party contract manufacturers for production of its product candidates and companion diagnostics.
