In the company’s own words
Replimune Group, Inc. (NASDAQ: REPL) is a clinical-stage biotechnology company focused on developing oncolytic immunotherapies designed to transform cancer treatment by maximizing immune system activation against tumors. The company's proprietary RPx platform is based on an engineered strain of herpes simplex virus 1 with multiple therapeutic payloads intended to achieve dual local and systemic anti-tumor activity through direct virus-mediated tumor cell killing and the induction of durable systemic immune responses.
The company's lead product candidate, RP1 (vusolimogene oderparepvec), is a selectively replicating HSV-1 virus expressing GALV-GP R(-) and human GM-CSF. RP1 has been evaluated in the IGNYTE clinical trial for advanced melanoma, demonstrating a confirmed overall response rate of 32.9 percent when combined with nivolumab in anti-PD-1 failed patients, with a median overall survival of 32.9 months and three-year survival rates of 47.8 percent across all treated patients. The company submitted a Biologics License Application to the FDA in November 2024 under the accelerated approval pathway and received Breakthrough Therapy designation. Following two Complete Response Letters, the company resubmitted the BLA in 2025 with an FDA action date of August 2, 2026.
Pipeline candidates include RP2 (sturlimgene erparepvec), engineered to express an anti-CTLA-4 antibody-like molecule for enhanced immune activation, currently in development for metastatic uveal melanoma through the REVEAL Phase 2/3 trial. RP3, designed with additional immune co-stimulatory pathways, is not currently being pursued due to program prioritization.
Replimune operates a dedicated in-house manufacturing facility in Framingham, Massachusetts, spanning approximately 63,000 square feet and designed to support full global commercialization. The company collaborates with Bristol Myers Squibb, which provides nivolumab at no cost for clinical trials, and maintains a signal-finding collaboration with Roche for hepatocellular carcinoma studies. The company's approach combines multiple mechanisms of action into off-the-shelf therapies intended to work synergistically with checkpoint inhibitor therapies and other cancer treatments.
