What PLIANT THERAPEUTICS, INC. does
In the company’s own words
Pliant Therapeutics, Inc. (NASDAQ: PLRX) is a clinical-stage biopharmaceutical company focused on discovering and developing integrin-based therapeutics. The company's website is www.plianttx.com.
Pliant's lead product candidate, PLN-101095, is an oral small molecule dual selective inhibitor of αvβ8 and αvβ1 integrins in development for treating solid tumors resistant to immune checkpoint inhibitors. The company's approach targets transforming growth factor-beta (TGF-β) activation through integrin inhibition, with the goal of sensitizing tumors to checkpoint inhibitor therapies in heavily pretreated patient populations. In 2025, Pliant announced positive Phase 1 data demonstrating anti-tumor activity when PLN-101095 was combined with pembrolizumab, with clinical responses observed across multiple tumor types including cholangiocarcinoma, melanoma, head and neck squamous cell carcinoma, and non-small cell lung cancer.
The company has assembled a proprietary drug discovery platform consisting of a library containing over 15,000 integrin binding molecules, complemented by comprehensive screening assays and an advanced live human tissue program. Beyond its lead oncology program, Pliant is exploring integrin-based target-mediated drug delivery applications, with current early-stage programs focused on delivering siRNAs to skeletal muscle cells, adipocytes, and renal cells across multiple disease areas.
Pliant previously developed bexotegrast, an oral small molecule dual-selective inhibitor of αvβ6 and αvβ1 integrins, for idiopathic pulmonary fibrosis and primary sclerosing cholangitis. The company discontinued the Phase 2b BEACON-IPF trial in 2025 following an independent data safety monitoring board recommendation, though early efficacy signals were observed. The company continues evaluating development pathways for the PSC indication.
The company's intellectual property portfolio includes over 300 pending patent applications worldwide across more than 30 patent families, with fourteen U.S. patents and forty-four foreign patents issued as of March 2026. Patents are expected to expire between 2037 and 2047. All clinical manufacturing is outsourced to third-party contract manufacturing organizations, with drug substance supply established from Asian CMOs to support both clinical and preclinical requirements.
